Mode of hemolytic action of the antifungal polyene antibiotic filipin.

چکیده

Filipin at low concentrations rapidly hemolyzes bovine erythrocytes. Of the compounds tested, which were known a constituents of cell membranes, only cholesterol and phosphatidyl-choline prevented hemolysis. A specific complex was formed only with cholesterol. Filipin treatment caused a release of cholesterol from erythrocytes. Since all cell sterol is located in the cell membrane, the hemolytic action of filipin may be due to a displacement of cholesterol from the cell membrane thus weakening its stability. The internal pressure of the cell then could force the hemoglobin through the defective membrane.

Effects of four components of the polyene antibiotic, filipin, on phospholipid spherules (liposomes) and erythrocytes.

Effects of a complex polyene antibiotic, filipin, and its four components were studied upon biological (erythrocyte) and artificial (liposome) membranes in order to test the “sterol receptor” hypothesis of polyene action. The hemolytic activity of filipin complex could be removed more effectively by liposomes which had cholesterol incorporated (phosphatidyl choline (7)~dicetylphosphate (2)-chol...

Steroid interference with antifungal activity of polyene antibiotics.

Wide differences exist among the polyene antibiotics, nystatin, rimocidin, filipin, pimaricin, and amphotericin B, with reference to steroid interference with their antifungal activities against Candida albicans. Of the numerous steroids tested, ergosterol was the only one which effectively antagonized the antifungal activity of all five polyene antibiotics. The antifungal activities of nystati...

Natamycin inhibits vacuole fusion at the priming phase via a specific interaction with ergosterol.

The antifungal antibiotic natamycin belongs to the family of polyene antibiotics. Its antifungal activity arises via a specific interaction with ergosterol in the plasma membrane (te Welscher et al., J. Biol. Chem. 283:6393-6401, 2008). However, this activity does not involve disruption of the membrane barrier function, a well-known property of other members of the polyene antibiotic family, su...

Effect of fatty acids on action of polyene antibiotics.

Fatty acids cause a decrease in the absorption spectra of the antifungal polyene macrolide antibiotics nystatin, filipin, candicidin, and amphotericin B. For nystatin, filipin, and candicidin, this decrease in absorption could be correlated with the activity of the fatty acids in protecting the yeast Saccharomyces cerevisiae against the action of these antibiotics. With amphotericin B a correla...

The Effect of Polyene Antibiotic Filipin on Pig Red Blood Cells

Polyene antibiotic filipin (Fil) is a potent hemolytic agent. Our previous kinetics analysis showed that the mechanism of Fil action on human erythrocytes can be classified as the damage type. Results of other experiments performed by us demonstrated that the kinetics of hemolysis induced in RBCs by two other polyene antibiotics, namely amphotericin B (AmB) and nystatin (Nys), is more complex, ...
